A two-letter word is the shortest you can play in Scrabble, and you’ve actually got several options if you want to go this route with only vowels.
Bonus: Because these words with only vowels are so short, they’re some of the easiest to memorize.
AA: This type of jagged basaltic rock will get you street cred with any geologists you happen to be playing.
AE: This short Scottish word means “one.”
AI: Not only is this word short, it’s also slow: it’s a South American sloth.
OE: Play this as an abbreviation for “Old English.”
OI: This is a Yiddish expression and also a subgenre of punk rock.
If you’re playing in the US, you have one longer Scrabble word with only vowels. EAU is French for “water,” and you might have heard it used in the names of perfumes or colognes.
If you’re playing using the UK Scrabble dictionary, you actually have a ton more options. These are all playable all-vowel words according to UK rules:

EUOI: In ancient Bacchic revels, this was a cry of happiness.
EUOUAE: This is a cadence in certain types of music.
Does Y count as a vowel? If it does, then you’ve got even more playable words with only vowels.
Bonus: Because Y is worth four points, these tend to get you more points overall than other vowel-only Scrabble words.
AY: This is interjection of sorrow or distress.
OY: This is an alternate spelling of the Yiddish expression for frustration.
YA: Especially in literature, this is an abbreviation for “young adult.”
